PREDICTION: This episode with either end with (a) Applebloom getting her cutie mark for real (probably not) or (b) Sweetie Belle and Scoots become inflicted with the disease once Applebloom gets over it and everyone laughs (except for Sweetie and Scoots).

The cutie pox as chicken pox angle works best for Applebloom and the rest of the CMC.
Perhaps one of the main six never got it, and we could see an adult become spotted with extra cutie marks?

The show wouldn't need to delve into the more serious effects adults can experience with late in life infection. (Shingles ->shingle -> plank -> plank flank?)

Although I don't expect the episode to take that direction, if it does I will feel for the pony who got late cutie-mark pox. Shingles are terrible, and the thought of them brings back memories of the time I had them for a week.

The potentially harsher effects of adulthood infection is what motivates the practice of putting other children in close proximity with a child that has it, so that they can get it out of the way.
(Unfortunately, since varicella-zoster is a life-long infection, parents were making a trade-off between the dangers of a full-blown chickenpox infection in adulthood versus the possibility of late-life shingles. The more you know...)

The tradition could be a motivation for getting the other fillies into the episode, if it is applied in the show. I'm not sure that the show would do that, Applebloom's getting at least one cutie mark would be enough to draw in the rest of the CMC.

I have fuzzy recollection of other shows with children that probably had chicken pox episodes.
A predictable plot twist would be an adult who suddenly remembers he or she didn't have it as a child after being exposed.

With how crazy they seem to be going drawing interesting new backgrounds for every single shot this season, I bet they're just itching to draw some awesome street scenes in Canterlot. It'll be like when all us Tolkien nerds first saw the inside of Minas Tirith on the big screen.

(Someone (Scuderia) should do a tally of all the unique backgrounds in a few sample episodes and see if they're actually getting more complex about it, or if it's just my imagination because the new stuff has all been so interesting and unique so far. Sweet goddamn I want to have a picnic in that spot where they had it in Lesson Zero.)

Even more spoilers: After Rarity is written out of the show, Lyr- I mean Heartstrings will spend five or so episodes being the sixth friend, but then they will fire her, claiming her to be "lame", so they're going to hold an competition for who's going to be the sixth friend. Heartstrings, meanwhile, will team up with one of the ponies that got eliminated from the competition and form a supervillian duo. Unfortunately they suck at it because they're aren't evil at all.

In the end they get a new friend, a completely new character named Coffeebeans, who is always highstrung due to all the coffee she drinks. This will go on for maybe five episodes before she disappears completely, as if the whole thing never happened.

Rarity eventually returns in the end of the season finale with no explanation. However she'll do less and less in the show except for maybe one or two episodes per season and occasional dialogues. Heartstrings, meanwhile, will continue to be prominent in the show in the future episodes, eventually becoming popular on her own right...
